What Is the Tesamorelin + Ipamorelin Blend?
This blend combines one peptide from each of the two main classes used in growth hormone axis research. Tesamorelin is a GHRH analog: the full 44-amino-acid GHRH sequence with a trans-3-hexenoyl group attached to the tyrosine at position 1. That cap blocks the enzyme DPP-IV, which normally breaks GHRH down within minutes. Tesamorelin is the active ingredient of the FDA-approved drug Egrifta. Ipamorelin is a growth hormone secretagogue that mimics ghrelin. It was developed by Novo Nordisk in the 1990s and described in early studies as the first selective secretagogue: it triggered growth hormone release without significantly raising ACTH or cortisol, unlike older peptides such as GHRP-6. Because GHRH-receptor and ghrelin-receptor signals reach the same pituitary cells through separate routes, researchers often study one agonist from each class together. This blend supplies both from one lot, under one certificate of analysis. The combination itself has not been studied as a single formulation in published research; each peptide’s literature stands on its own.

Tesamorelin and Ipamorelin in Published Research
The two components have very different research records. Tesamorelin went through full FDA registration trials before Egrifta was approved in 2010, so its effects on growth hormone release, IGF-1 levels and visceral fat tissue are documented in large, placebo-controlled human studies. Later research has also examined it in liver fat and cognitive studies.
Ipamorelin’s record is mostly preclinical, with some early human work. Studies in rats characterized its selectivity for growth hormone release, and it reached Phase 2 human trials for a gut-motility indication before development stopped. It was never approved. On October 29, 2024, an FDA advisory panel voted 0 to 12, with 1 abstention, against adding Ipamorelin to the compounding pharmacy list.
